The US Food and Drug Administration (FDA) is the first regulatory body to begin to use miniature models of a human organ โ€“ the liver. These models (pictured) are made from different types of human liver cells grown on a scaffold. This scaffold has blood-like fluid pumped through it to deliver nutrients and remove waste products. The FDA want to find out if these โ€˜livers-on-chipsโ€™ can reliably model how our bodies react to new foods, including added colours and flavours, and food-borne illnesses. If its initial trial is successful, the FDA may start testing on kidney, lung and intestine models. These could more closely replicate how the cells in our body behave than is currently possible with computer models and through studies on human tissues and animals. And ultimately reduce the number of animals used in testing. However, it will still be some time before โ€˜organs-on-chipsโ€™ replace traditional research techniques.

No Default Sex

Male and female embryos start out roughly the same, but as development progresses, the tissues that will ultimately become sperm- or egg-producing structures are either retained or eliminated, depending on sex. It was thought that genes present only on the Y chromosome controlled the development of male-specific structures, and that the lack of a Y chromosome resulted in female-specific structures. In other words, that male development was an active process and female development the default. It is now clear, however, that the development of female sex organs is active too. Researchers have discovered a factor produced in female mice necessary for the elimination of male-specific tissues. The mutant embryo (bottom) lacks this factor and thus retains both female and male precursor tissues (pink and blue). This new discovery should help to inform studies into both intersex development and certain reproductive problems.

Found in every tissue in the body, macrophages are large immune cells known for their ability to engulf and destroy pathogens, yet they also perform many other duties. In the testes, they are involved in both its key functions, the production of sperm and testosterone. Pictured in a mouse testis, macrophages (in turquoise) surround the seminiferous tubules (in red), where sperm are made. As sperm are only produced after puberty, other immune system cells may see them as โ€˜foreignโ€™ tissue so testicular macrophages also secrete defensive proteins to protect them. Two types of macrophages, with different developmental pathways, fulfil these crucial roles. Those found alongside testosterone-producing Leydig cells in the interstitial spaces of the testes originate from precursors in the embryo, while macrophages around the seminiferous tubules develop at puberty, from bone marrow cells. A deeper understanding of both types of macrophages could prove relevant to studies of male fertility.
